The outbreak of Zika virus in 2016 made scientists start to look for new alternatives for detection of this virus. Although methods like reverse transcription polymerase chain reaction (RT-PCR) or enzyme-linked immunosorbent assay (ELISA) have been the most utilized methodologies, and have commercial versions for Zika virus detection, they are not enough for detection in the case of a mass infection, once the high demand and necessity of an advanced structure for detection utilizing these methods are necessary. Surface enhanced Raman spectroscopy (SERS), on the other hand, has been utilized in analytical chemistry, as well as in bioanalytical, having the capacity of detection in picomolar concentration and distinguish samples, using the spectral fingerprint, becoming an alternative for Zika virus detection. This way, we were able to ally SERS, utilizing gold nanoparticles (AuNPs), and the physical preconcentration by superparamagnetic iron nanoparticles (SPIONs), to characterize and detect Zika virus antibody or NS1 Zika virus protein, as can be seen in Figure 1. It was possible to characterize Zika virus antibody and NS1 protein utilizing SERS, and in the concentration of 1µg/L (about 10 pM), opening a new expectation for the uses of this method to developing of a sensor of Zika virus using Raman spectroscopy, directly, label-free.
