Our lab recently reported copper probes that undergo a copper-catalyzed oxidative cyclization of azo-anilines to benzotriazoles. Such chemical conversion is accompanied with both dramatic color and fluorescence changes so that these signal transductions can be used to determine residual level of copper in the sample. By enriching electron density on the copper probe, we demonstrated that this colorimetric/fluorimetric method can be completed in less than 1 minute, and can detect very low amount of copper (ppm level) in pharmaceutical compounds. As real-world applications, our copper probe was coupled with the other analytical techniques such as HPLC (high performance liquid chromatography) and MS (mass spectrometer) to realize automated assay of multiple samples such as 96-well plate. In this presentation, we will discuss the development of optimal copper probe, and three different ways to apply this probe to actual pharmaceutical process development; i) MISER (Multi injections in a single experimental run), ii) flow-injection mode, and iii) real-time monitoring of copper removal cartridge.
