Pantetheine is a prosthetic group which is post-translationally attached onto the acyl carrier protein (ACP), or central carrier protein, within fatty acid and polyketide biosynthesis. The interactions of pantetheine with ACP are significant, as they dictate how the carrier protein interacts with the growing fatty acid or polyketide chain en route to interacting with partner proteins. Pantetheinamide mimetic probes have been shown to capture key interactions of <i>E.coli</i> ACP, AcpP, in these biosynthetic pathways. These interactions are important in understanding how this ‘arm’ interacts with AcpP during these biosyntheses and how it influences chain sequestration, or the protection of unstable intermediates within a hydrophobic pocket prior to interaction with its partner proteins. Herein, to better identify how these pantetheinamide interactions manifest within ACP systems, we have developed a route to site specifically labeled probes with <sup>15</sup>N isotopes for studying these key interactions with AcpP.
