Kratom is an evergreen tree in the coffee family native to Southeast Asian that has been used for traditional medicine to relieve pain. The primary active alkaloid, mitragynine, found in Kratom leaves affects the same receptor as the opioids, such as heroine, morphine, and codeine. The purpose of this study is to develop an efficient solid phase extraction method for mitragynine from dried Kratom powder.<br/><br/>Oasis MCX cartridges were used which are of a mixed mode. They have a strong cation exchange and are reversed phase. These cartridges are a water wettable polymer which are selective with bases and stable in organic solvents. The instrument for analysis is a Shimaduzu HPLC with an OCTYL 5µm 250mm x 4.6mm column. The sample was eluted isocratically with a mobile phase of 0.05% formic acid and acetonitrile at pH 5.0 (50:50, v/v), flow rate = 1.0 mL/min. Mitragynine was detected at 223 nm with a retention time of 2.0 min. A calibration curve was prepared with mitragynine standards ranging in concentration from 0.00626 mg/mL to 0.500 mg/mL. Linear correlation was excellent with a Pearson correlation coefficient of 0.9996 which is above 95% confidence and an R&lt;span style="font-size:10.8333px"&gt; &lt;/span&gt;squared value of 0.9993. The amount of mitragynine in the Maeng Da strain was determined to be 12.25 mg/g. Intra-day study shows an RSD of 5.813 % (n =3).
