Extracellular vesicles have been a growing area of research partly due to the concept of liquid biopsies, or the analysis of tissue in fluids without the need of invasive surgery.  Asymmetrical flow field flow fraction (AF4) has many benefits including; high recovery, easy buffer exchange, gentle size separation, and the ability to wash vesicles as they are isolated.  By utilizing the size separation and washing effects AF4 provides, immunolabeling and membrane dyeing can be achieved during focusing of the sample in the channel during separation, thus dramatically speeding up the downstream protein analysis commonly performed after isolation.  High recoveries reduce sample input while buffer exchanges to volatile buffer system allows for salt free purification of samples, making this an ideal system for routine vesicle analysis in cell culture medium.<br/>
