Single nanoparticle near-infrared surface plasmon resonance imaging (SPRI) microscopy is a label-free spectroscopic method for measuring the integrated refractive index of individual  nanoparticles. The adsorption of a hydrogel nanoparticle onto a chemically modified gold thin film creates a point diffraction patterns int the SPRI differential reflectivity images. The intensity of this diffraction pattern can be quantitated to create a single nanoparticle reflectivity change value, Δ%Rnp. We have demonstrated the that the distribution of Δ%Rnp values from a set of hydrogel nanoparticles can be used to obtain size and content information.  We have synthesized and characterized various hydrogel nanoparticles that have incorporated short single-stranded DNA (ssDNA) and ferrite magnetic nanoparticles, and we have demonstrated how to use these hydrogel nanoparticles to measure bioaffinity uptake and release.
