The application of electrochemical, aptamer-based sensors for application in, for example, wearable and implantable sensors requires that the sensor response be stable over the lifetime of the deployment of the sensor. The sensing mechanism of this class of sensors relies on a specific target-induced conformation change of an electrode-bound aptamer recognition element. This specific interaction combined with faradaic electrochemical readout typically leads to robust sensor performance in complex environments. However, the sensor platform is not impervious to long-term signal drift on account of nonspecific binding of biologicals over the minutes-to-hours time scale. In this talk, I will discuss our recent results on the use of nanoporous gold leaf electrodes to improve the long-term stability and stable base-line performance of electrochemical, aptamer-based sensor. We find that the use of these nanoporous gold films, when employed as the sensing working electrode, impeded long-term drift and provide stable sensor performance in 100% undiluted blood serum and whole blood without compromising overall analytical performance of the sensor. The presentation will cover the fabrication, characterization, and application of this type of sensor.<br/>
