It is recognized that drug and gene delivery systems have numerous advantages for controlled drug release, including prolonged duration time, reduced side effect, improved drug bioactivity and enhanced therapeutic efficiency. To enhance the bioavailability of therapeutics and to deliver therapeutic agents to particular tissues and cells, the ideal drug and gene delivery systems should have desired functional properties, including target ability and stimuli responsibility. Our studies focus on the functional peptides for biomedical applications. A series of functional peptides and fusion peptides with particular functions, such as tumor cell targeting, organelle (nucleus and mitochondrion) targeting, endo/lysosomal escape and cell penetrating, were synthesized. Various tumor targeting delivery systems with synergistic association of diverse responsibilities based on peptides and hybrid materials containing peptides were constructed for drug and gene delivery. The delivery capability and anti-tumor efficiency of these delivery systems were evaluated. Both in vitro and in vivo studies show these systems can exert therapeutic actions and inhibit tumor growth efficiently through delivering the therapeutic agents to tumor cells with high efficacy. In addition, different types of functional nanoparticles containing functional peptides for theranostics and multimodal therapeutics were constructed to overcome the critical issues in tumor treatments.
