Osteoporosis is a highly worldwide disease caused by reduction of bone mass and microarchitectural deterioration, leading to bone fragility and fracture. Early diagnosis of osteoporosis is the key subject for efficient identification and therapy of patients with high risk of fractures. In this work, we have demonstrated a platform for rapid and effective detection of bone biomarkers by surface-assisted laser desorption/ionization mass spectrometry (SALDI-MS) based on two-dimensional multilayered gold-nanoparticle thin film (MTF-AuNPs). With strong surface plasmon resonance and excellent homogeneity, facilely-prepared, highly-ordered and large-scale MTF-AuNPs sample substrate revealed high sensitivity of bone biomarker of hydroxyproline (HYP) in SALDI-MS measurements without additional matrixes. Unlike other matrixes commonly used in SALDI-MS that required numerous sample preparation procedures; on contrast, by a simple one-step deposition of analytes onto the sample substrates of MTF-AuNPs, the detections were ready to perform. The excellent reproducibility of HYP spectra with only 9.3% relative signal variation could be attributed to MTF-AuNPs’ high absorbance at wavelength of 337 nm, low heat capacity, superior thermal conductivity and outstanding homogeneity. Furthermore, the serum sample was directly deposited onto the MTF-AuNPs sample substrate without any pretreatment and its HYP concentration was then successfully determined. Our results showed that the signal contamination, signal-to-noise ratio and reproducibility of mass spectra with the use of MTF-AuNPs sample substrate have been significantly improved in comparison to the uses of commercial CHCA or DHB as the assisted matrixes.
