Chemical tools and technologies are continuing to shape our understanding of biomolecules.  Included in this group are chemical reporters—unique functional groups that can be incorporated into diverse biomolecules and detected with highly selective (i.e., bioorthogonal) reactions.  Despite their myriad successes, many chemical reporters remain too large or insufficiently stable for routine cellular use.  We recently developed one bioorthogonal reagent—cyclopropenone—as a general and versatile chemical reporter.  Cyclopropenones are small, biocompatible functional groups that exhibit unique manifolds of reactivity.  Not only are these motifs suitable for biomolecule tagging (via bioorthogonal ligation with phosphines), they can also be used to control biomolecule function via phosphine-mediated crosslinking.  Cyclopropenones are further compatible with many popular bioorthogonal cycloaddition reagents, enabling multicomponent labeling studies.  The development and applications of bioorthogonal cyclopropenones will be discussed, with an emphasis on targeting and activating cellular proteins.<br/>
