Helicobacter. Pylori (H. Pylori, HP) is a leading cause of common human gastroduodenal diseases such as gastritis, peptic ulcer, and gastric cancer. The formation of biofilm increases H. pylori resistance to adverse environments. a number of treatment failures of HP-related infections arise from the protective environment of biofilm. Previous studies have suggested that curcumin (CCM) inhibits the growth and biofilm formation of HP. However, the low solubility of CCM limits its bioaccessibility and anti-HP efficacies. The objective of this study is to evaluate the capabilities of octenylsuccinate hydroxypropyl phytoglycogen (OHPP), a recently developed amphiphilic dendrimer-like biopolymer, to enhance the aqueous solubility of CCM and its inhibitory efficacy against HP biofilms. In this study, a solid dispersion was prepared through incorporating CCM with OHPP using co-solvent mixing followed with spray-drying at the CCM/OHPP mass ratio of 1/4. The solubility of CCM in CCM-OHPP solid dispersion was over 5700 times that of pure CCM. In the dissolution test, around 47% of CCM was released from the solid dispersion within 20 min, whereas only 0.3% of CCM was released from pure CCM. Compared with untreated HP biofilm, CCM-OHPP treated HP biofilms showed lower amount of extracellular polymeric substances, fibrillar networks, and complex bacterial layers. We expect that CCM released from CCM-OHPP solid dispersion will inhibit HP biofilm in a dose-dependent manner.
