Silk fibroin is a naturally sourced, biocompatible material that lends itself to in vivo applications as a biomaterial. Here, we report two methods of covalent modification on silk fibroin via tyrosine residues. We demonstrate the attachment of alkynes and azides and subsequent biorthogonal click reactions with these newly installed functional groups. Synthesis methods as well as characterization using 1H NMR, 1H-15N Heteronuclear Multiple Bond Correlation (HMBC) NMR, ATR-FTIR and UV-VIS will be presented.
