Recently, mass coral bleaching has occurred in virtue of factors such as temperature, radiation and pathogenic microorganisms, which results in changes of coral community structure and degradation of global coral reef ecosystem. In order to realize the sustainable conservation and management of corals, it is urgent to design intelligent bio-materials as culture medium simulating native micro-environment accurately. However, instructing cell adhesion at the interface between synthetic materials and the biological environment remains a tremendous challenge. Herein, composed of RGD peptide modified carboxymethyl chitosan (CCS-R) and dopamine modified oxidized sodium alginate (OALG-D), the fabrication of multilayered CCS-R/OALG-D film is conducted with via layer-by-layer (LbL) self-assembly based on Schiff base reaction. The film possesses high adhesion to cells, which is beneficial to cell growth. Furthermore, it holds the excellent self-healing, biocompatibility and antioxidant properties. This self-healing film, as an extracellular culture matrix, has prolonged the survival of coral explants <i>in vitro</i> owing to its super cell adhesion. Accordingly, the integration of self-healing, high adhesion, superior cell adhesion, and antioxidant properties into a facile LbL self-assembly film has been expected to open a promising window for the application of simply prepared two-dimensional materials to the marine conservation.
