A series of ruthenium(II) complexes will be presented that are able to both generate singlet oxygen in high yield upon irradiation and also undergo photoinduced drug release.  New complexes will be presented that release nitrile- and pyridine-based inhibitors that target enzymes overexpressed in cancer, as well as strategies to increase tumor localization of the agents.  The ability of the complexes to also generate singlet oxygen to effect cell death signficiantly increase their activity against a number of cancer cell lines and 3D tumor models. Ligand platforms to enhance the absorption of red and near-IR light will be discussed.
