Multiple anti-cancer compounds were tested to determine their cytotoxic activity against both cancer and non-cancerous cells. Based on the Selectivity Cytotoxicity Index, one compound, NC 2559 was selected for further research. Different assays were performed to determine which cell death mechanism was caused by compound NC2559. Both the Annexin V assay and Caspase 3 assays indicated that the compound induces apoptosis. In addition, NC2559 induces Reactive Oxygen species and mitochondrial membrane depolarization. Two genes, HMOX and PMAIP1 (NOXA) were found to be highly induced by RT-qPCR that are related to oxidative stress. These data demonstrate that NC2559 induces apoptosis in acute promyelocytic leukemia cells via the intrinsic pathway.
