Interlocked molecules, like rotaxanes, are typically synthesized by threading a linear axle-like molecule through a macrocyclic host. The thread is then held in place by large molecular caps that are coupled to the ends of the threads. This is a viable synthetic method we have previously employed using various macrocyclic hosts and subsequent linear guests. This work focuses on the design and synthesis of cucurbit[6]uril (CB6) based rotaxanes containing pH-sensitive cleavable axles. The aim is to use these cleavable CB6-based rotaxanes as novel biosensors for the early detection of lung cancer.
