The applicability and/or generality of molecular features and how they impact small molecule permeation into Gram-negative bacteria remains poorly understood. Identifying motifs or structural properties that correlate with permeation would enable the rational design of novel broad- and narrow-spectrum antibacterials. Herein we highlight the results of an investigative study on a strategically designed library aimed at revealing the effects of molecular structure on Gram-negative accumulation and biological activity. <i>E. coli, A. baumannii</i>, and <i>P. aeruginosa </i>strains exhibiting a spectrum of efflux and outer membrane competencies were leveraged to identify motifs that pose liabilities to outer membrane permeation and efflux in broad and species specific manner.
