As a key player in anti-tumor immunity, CD8+ T cells recognize tumor-associated antigens and kill cancer cells. Upon activation, naive CD8+ T cells undergo rapid cell division and differentiate into effective T cells which process is supported by extensive metabolic rewiring. To understand the metabolic reprogramming during T cell activation, we applied untargeted metabolomic analysis to compare in vitro activated CD8+ T cells with their naive counterpart. With our in-house developed software package, we performed time-course analysis and identified novel metabolic markers that are associated with T cell receptor (TCR) signaling independent of nutrient environment. Those associated metabolic pathways were validated in vivo to be important for T cell proliferation and their cytotoxic functions.
