Amide derivatives of rhodamine B have been used in a variety of sensing applications like visual indicators and pH-based sensors due to the on/off responses of its amides to stimuli, such as acidity changes and UV light. In collaboration with the Lundin lab at High Point University, we synthesized amide-linked dimers of rhodamine B, which were shown to fluoresce two different colors, orange and blue, depending on the equilibrium between the open, amide form, and a closed, spirolactam form. Change in the temperature of amide-linked rhodamine B influences a shift in equilibrium between its orange and blue states. In fluorometry, the excitation and emission wavelengths of these substances were collected from 3D fluorescence spectra at varying temperatures to determine different thermodynamic factors associated with the rhodamine B dimer which will further aid in potential methods of control.
