Higher carbon sugars, such as nonulosonic acids, are known to be important structural components of oligosaccharides found on the surfaces of human cells, bacteria, fungi, and other pathogenic and nonpathogenic organisms. Nonulosonic acids have been synthesized from six-carbon precursors by chemoenzymatic pathways utilizing aldolases from <i>E. coli</i> and <i>Pasteurella multocida.</i> Various other organisms, such as hyperthermophiles, also possess enzymes that catalyze such aldol reactions. Herein we present efforts to develop the first high throughput UPLC-MS assay to screen aldolases with nonnatural substrates to aid in the use of these enzymes for carbohydrate building block synthesis.
