Antibiotic resistance in bacteria is a growing problem, necessitating the development of potent new antibacterial compounds. The T-box riboswitch is an RNA drug target that is found in Gram-positive bacteria. The NMR-derived solution structure of the T-box riboswitch antiterminator has been elucidated, which allows for in silico docking studies to be performed. Additionally, a linear correlation has been observed between these in silico docking studies and Kd values determined in RNA-binding assays involving the T-box riboswitch. This allows for docking studies to be performed, with the potential for RNA-binding ligands to be identified. We will report on the in silico docking studies, as well as the synthesis of novel RNA-binding ligands.
