Hydrophilic EGCG and lipophilic lycopene was co-delivered within a water-in-oil-in-water (W1/O/W2) double emulsion system for the enhanced stability and synergistic hypolipidemic effect. To conquer the inherent instability issue of double emulsion system, a Pickering type double emulsion formulation (PDE) was introduced via utilizing bacterial cellulose (1% w/v) as the external interface stabilizer. EGCG and lycopene, two naturally occurring phytochemicals with extreme sensitivity to processing and gastrointestinal environments, were encapsulated in the internal water and oil phase, respectively. The structural evolution profile of PDE during storage and in vitro digestion, the antioxidant properties of lipid phase during storage, the retention rate and the release profile of EGCG and lycopene during storage and digestion were systematically investigated. Furthermore, the synergistic hypolipidemic effect of the co-delivery system was verified in a high-fat diet rat model. PDE fabricated under the optimized compositions of 15% v/v W1 (0.23 mg/mL EGCG), 35% v/v oil (0.23 mg/mL lycopene) exhibited structural stability during 14 days’ storage. The EE% of PDE fabricated via the two-step emulsification method retained 97.31 % EGCG within the internal phase. The retention rate of co-encapsulated EGCG and lycopene during two weeks’ storage was 3.2 and 1.7 times higher than that of free EGCG and lycopene solutions, respectively. Encapsulation of EGCG and lycopene within PDE not only resulted in higher chemical stability but also increased their bioaccessibility significantly in simulated gastrointestinal fluids, a 9.1-fold and 4.7-fold augmentation in their bio-accessibility was achieved. The TC, TG, HDL-C, LDL-C contents in the blood of rat model with dyslipidemia after 30 days’ treatment suggested that EGCG and lycopene co-loaded in PDE demonstrating synergistic effect against dyslipidemia. PDE is a promising technology for co-loaded hydrophilic and lipophilic bioactive substances with enhanced stability and synergistic health beneficial effect.
