<br/>Cisplatin is one of the first line drug for the treatment of various cancer such as testicular, ovarian, cervical, head and neck cancer but it’s used has been limited clinically due to severe side effect such as nephrotoxicity. Therefore, it is very important to design probes and strategies to monitor the onset of nephrotoxicity for better treatment. Using an iridium complex, we were able to monitor the progress of cisplatin induced kidney injury by monitoring the level of superoxide anion produced. Cisplatin induced kidney injury is characterize by the production of increase level of reactive oxygen species, with superoxide anion being the primary ROS produced. This probe shows excellent selectivity and sensitivity towards superoxide anion over a broad range of pH and over other reactive oxygen species. This probe was successfully applied to detect endogenously produced superoxide in living cells and to detect superoxide in fetal bovine serum (FBS). Additionally, this probe can be efficiently applied to visualized cisplatin induced kidney injury in mice and also to examine the protective effect of L-carnitine in cisplatin induced kidney injury. It is noteworthy that this the first time a metal complex in applied in the detection of superoxide in FBS and to visualize cisplatin induced AKI in mice. We believe this work will open up great avenue for exploring metal complexes for unmasking the role of superoxide in nephrotoxicity and other health related diseases.
