Neurotrophins are proteins facilitate the growth and development of neurons. While neurotrophins have significant potential to combat neurodegenerative diseases, they do not efficiently cross the blood-brain barrier and have poor half-lives in serum. Many small non-peptidic molecules have been identified to elicit similar phenotypic responses (neurotrophic small molecules). Phenylbutenoid dimers are two such compounds, originally isolated from <i>Zingiber Purpureum</i>, commonly called Javanese ginger, and show neurotrophic and neurogenic effects in both cell culture and mouse studies. However, the molecules have poor potency (~ 30 μM), and a mechanism of action or binding target has not yet been elucidated. Our research aims to investigate the structure-activity relationship for these neurotrophic molecules in order to develop more potent analogs, and also facilitate elucidation of mechanism of these neurotrophic molecules. Such information may provide insight for new medicinal strategies for neurodegenerative diseases.
