The Rossi number (Rn) is a pure number capable to characterize the deformation of a liquid film of Fomblin HC-25 (PFPEf) when a liquid system come in contact with its surface. Rn is measurable by the contact angle method. The aim of our investigation was the evaluation of the applicability of Rn on the permeation process of carnitine and colic acid released from a test ointment across an ex vivo biological membrane, performed by Franz’s cell. The quantitative analysis of carnitine and colic acid was carried out by a HPLC apparatus equipped with an Evaporative Light Scattering detector. Rn was carried out by contact angle analysis of 80 µl of acceptor solutions collected from each of 12 Franz’s cells used for the assay, at different times from 3h to 12h. The results demonstrated the applicability of Rn on the analysis of the variation of the concentration of active substances over the time at 3, 6 and 12 hours (t3=9.07±1.4, t6=11.96±1.2, t12=14,23±1.4) and allow us to hypothesize that the deformation of the PFPEf liquid film it may be a new tool for the non invasive and rapid characterization of the permeation process in pharmaceutical technological field.
