A small library of dipeptidyl nitriles and their bioisosteres (sulfonamide based peptidyl nitriles) have been synthesized. The modifications at P2 and P3 have been made to study the structure activity relationship and obtain a selective Cathepsin B and L inhibitors. Further synthesis to modify P1 and synthesize more compounds with P2 and P3 adjustments are in progress. The synthesized compounds have been tested against a painel of four human cathepsins, CPB and cruzain enzymes.
