Olfactory receptors (ORs) are members of the seven-transmembrane
G-protein coupled receptor (GPCR) superfamily. Humans have a large number of OR genes (∼400 genes) most of which have been shown to be expressed in the nasal olfactory epithelium. Ectopically expression of OR genes in several non-olfactory
human tissues has also been described, although the physiological function has only been shown for a limited number of them. Previous studies have shown expression at the mRNA level in human keratinocytes for several ORs (OR6M1, OR11A1, OR5V1, and OR6V1). The expression of OR2AT4, OR2A4/7 and OR51B5 has also been previously demonstrated in human skin, and a possible role in wound healing has
been hypothesized for these receptors.
Herein we demonstrate expression for several additional ORs in human skin, both at the mRNA and the protein level. Our results have revealed different OR expression patterns in the epidermis of human skin. The expression levels for these ORs in cultured keratinocytes have also been explored. We have also observed qualitative differences in OR expression at the epithelial tongue in wounded skin with respect to nonwounded tissue. These findings support a role for olfactory receptors in skin homeostasis and/or wound healing and may represent promising targets for drug development.
