Our laboratory reported in 2014 on a water-soluble ruthenium compound containing an iminophosphorane ligand (Ru-IM, 1) as potential chemotherapeutic agent (J. Med. Chem.). The compound was found to be significantly cytotoxic to cisplatin resistant cell lines, while displaying a non-covalent interaction with DNA and a mode of action independent from p53 inhibition. Ru-IM (1) was especially promising in a highly aggressive and difficult to target triple-negative breast cancer cell line, MDA-MB-231, displaying high efficacy in vitro and in vivo. Here we report on the anti-migratory, anti-invasion and anti-angiogenic properties of Ru-IM (1) and we compare them to those of other Ru(II) and Ru(III) derivatives such as NAMI-A and RAPTA-C. 2D migration, 3D invasion, and angiogenic tube-formation assays will be discussed. In addition, we will summarize the effects of this compound on an NCI 60 cancer cell-line panel and we will show the effects on different triple-negative breast cancer cell lines. Furthermore, a proteomic assay to examine the effect of this compound on various protein markers of cancer proliferation in the triple-negative breast cancer cell line will be discussed. Finally, we will describe confocal microscopy assays on the effects of a luminescent analogue of Ru-IM (1) in the TNBC cell line MDA-MB-231. We expect to provide preliminary data to help elucidate the mode of action of this cheap, water-soluble and easy to prepare compound with a long shelf-life.
