Diamond nanoparticles provide outstanding chemical stability and can be modified to contain highly fluorescent Nv centers.  The high chemical stability of diamond provides the opportunity to study the influence of molecular and polymeric ligands on nanoparticle behavior and function. Using NMR and FTIR spectroscopy, we have been investigating the influence of ligand molecular structure on the properties and biological impact of nanoparticles.  Our results show that nanoparticles functionalized with polymers leads to loops and trains that extend out from the nanoparticle surface, increasing the nanoparticle's ability to interact with membranes and other nearby biological surfaces.  In contrast, shorter ligands, even with high positive zeta potential, exhibit much less impact on membranes and bacterials cells. These results show that while nanoparticle charge is one important factor controlling biological response, the specific spatial distribuion of charged sites is equally important.  The detection of nanoparticles in complex biological and environmental matrices can be greatly enhanced through the activation of nitrogen vacancy (Nv) centers, which are highly fluorescence defects buried within the nanoparticle core.  We will report recent results using the novel spin properties of Nv centers to enable new modalities of imaging of nanoparticles within complex matrices.
