Engineered colloidal nanoparticles (NPs) have shown huge promise in the applications in diagnostic, drug delivery, imaging and therapy. Currently, the experimental focus of assessing their impacts on living systems has been on particle design, since it is well known that the biological interactions of a nanoparticle are a function of its physicochemical properties, but not much on the biological system or microenvironments the NPs interacted with. Our lab have reported that the NPs indeed interact with the extracellular matrix (ECM) and interfere with cell behavior. We hypothesize that NPs in the ECM that surrounds cells alter local concentration gradients of some molecules and thereby influence cell behavior – without the need for the NPs to enter cells. Herein, we examine the cell-ECM-NP nexus for cases where the chemoattractant Monocyte Chemoattractant Protein-1 (MCP-1) in the media has a clear chemotactic effect on Monocytic Cell Line THP-1 cells to investigate the effect of different gold NPs on MCP-1 guided THP-1 chemotaxis. Cell movements are monitored via bright field optical microscopy in chemotaxis µ-Slide. The chemotactic effect of MCP-1 for THP-1 cells was reduced by PSS-AuNPs and the reduced chemotactic effect of MCP-1 for THP-1 cells was mitigated by purifying the PSS-AuNPs solution. The excess PSS in solution as well as the PSS wrapped on AuNPs can both affect the cell migration. While Citrate-AuNPs is negatively charged, the chemotactic effect was not significantly decreased. Neutrally charged PEG-AuNPs and positively charged PDADMAC-AuNPs has little effect on cell migration.<br/><br/>
