In this presentation, the hydrophobic subtraction model (HSM) and its associated database at hplccolumns.org are employed to show the advantages of using two shorter, different columns in series for the resolution of various sets of HSM and non-HSM compounds compared to using one longer column of equivalent plate number/peak capacity. With the aid of a computer program, results at pH 2.8 and pH 7 for the over 260,000 pairwise column combinationswere compared with those obtained for the &gt; 730 individual columns currently entered into the HSM database. Although there was a very small percentage of instances in which the best individual column result was better than any separation achieved with a pair of columns, in the overwhelming majority of cases there was a significant benefit to using two shorter, different columns in series. This can be explained by the selectivities achieved with the two columns in series, many of which are shown to be significantly different and superior to the selectivity provided by any of the over 730 individual columns in the HSM database.
