MicroRNAs (miRNAs) are a group of small non-coding single-stranded RNAs (~22nt) that mediate the translational repression. miRNAs regulate gene expression and play critical roles in multiple biological processes, including cell differentiation, proliferation, and apoptosis as well as disease processes such as cancer, Alzheimer’s disease, and myocardial disease. In particular, the role of miRNAs in cancers has been extensively studied. Therefore, miRNAs have been suggested as being useful in early diagnosis of many diseases and cancers. Frequently, the concentration of miRNAs are low in clinical samples and thus high sensitivity of the detection is required.<br/>Whereas Polymerase Chain Reaction (PCR), northern blotting, and microarray are standard methods to detect and quantify miRNA, the error in amplification process and low sensitivity are hurdles for the accurate analysis. In order to overcome the limitations of the conventional technology, we combined the microarray with a nano-scale controlled surface and metal-enhanced fluorescence (MEF) nanoparticles. To detect miRNA-134, we employed S9.6 antibody that is DNA/RNA hybrid-specific mouse monoclonal after allowing the binding of the specific miRNA, and the MEF nanoparticles tethering the secondary antibody. Because it is observed that the typical limit of detection is 1.0 fM, and the enhancement factor is more than 1,000 times.
