Artemisinin is a sesquiterpene lactone and a popular malaria drug in many parts of the world. Its mode of action in killing the <i>Plasmodium falciparum</i> parasite is not well understood. It is suggested that the endo-peroxide bridge found in artemisinin and its derivatives plays a key role in the overall mechanism of action. In this work, we investigated the reaction of the artemisinin and its derivatives with DNA bases in the presence of heme proteins. LC/MS was used to tract the reaction products and eventually establish the role of the heme enzymes in the activity of these derivatives toward DNA bases.  The results of key reaction products or DNA bases with artemisinin will be discussed.<br/>
