The fluorescence emission of 6-propionyl-2-dimethylaminonaphthalene (PRODAN) shows a large shift (more than 100 nm from cyclohexane to water) in solvents of varying polarity and hydrogen bonding abilities. This feature has made PRODAN an attractive fluorescent probe for studying heterogeneous biological systems. Despite its extensive use, the nature and number of PRODAN excited states remain a matter of debate. In this research, Fourier-transformed emission decay matrix (FT-EDM) excited in the UV and visible are measured in water, ethanol, octanol, acetonitrile and cyclohexane. The number of excited states contributing to each FT-EDM are estimated using a variety of rank estimation procedures. Further, non-negative matrix factorization (NMF), a self-modeling curve resolution algorithm, is used to extract the spectral and decay profiles of the probe excited states and investigate the solvent specific nature of the states that produce the decay matrices. Specifically, the photokinetic transfer matrix is calculated from the frequency-domain decays to elucidate the mechanism of excited state interactions in each solvent and detect any trends in those interactions as the polarity and hydrogen bonding ability of the solvent changes.
