Cell mimicry envisions to replace missing/lost cellular activity and to incorporate non-native function in mammalian cells and tissue.<br/>Our artificial cells and artificial organelles are based on bottom-up assembled reactors using polymeric building block in combination with biomolecules.<br/>We develop micellar-based artificial organelles with biocatalytic function using small organic compounds to circumvent the inherent instability of enzymes, with the goal to lower reactive oxygen stress in liver disease. In parallel, polymer-lipid hybrid vesicles were used for intracellular production of nitric oxide in primary human macrophages. In both cases, cytosolic placement of the assemblies is essential. Therefore, we are exploring polyanions with membranolytic properties to facilitate lysosomal escape of nanoreactors and/or their cargo.<br/>From a different perspective, artificial cells (micron-sized subcompartmentalized reactors) are explored as supporters for their biological role model. We illustrated that artificial cells can be employed for detoxification in hepatic cell aggregates, to kick-start mineralization in bone cell spheroids and to counteract certain aspects in exitotoxicity in neuronal communication.<br/>Taken together, artificial organelles and artificial cells, using polymeric building block in combination with lipids and proteins, are broadening the toolbox to address biomedical challenges.
