Platinum based chemotherapeutics have been the standard treatment for many different malignancies since the discovery of their antineoplastic properties by Rosenburg in 1969. Since then there has been a desire to produce organometallic chemotherapeutics with fewer negative side effects than platinum-based drugs exhibit. Ruthenium based chemotherapeutics have the potential to fill this need. The complex [Ru(Phen)2(imPhen)]2+ has been observerved in previous studies to show qualities similar to that of other candidates for chemotherapeutics. Our current research focuses on the synthesis, purification, and characterization of the complex. To make [Ru(Phen)2(imPhen)]2+, ruthenium(III) chloride is reacted with 1,10-phenanthroline (Phen) to produce [Ru(Phen)2Cl2]2+, which is then reacted with 1H-imidazo-[4,5-f][1,10]-phenanthroline (imPhen). The eventual goal of this study is to use atomic force microscopy (AFM) to observe the conformational changes of DNA upon complexation with [Ru(Phen)2(imPhen)]2+.
