In this study, we used fluorescence spectroscopy and NMR to study sequence specific binding of europium (III) and a europium (III): ciprofloxacin complex to DNA duplexes containing base pair mismatches. The fluoroquinolone ciprofloxacin was used to observe lanthanide binding via fluorescence. The europium (III): ciprofloxacin complex was titrated with short 10-13 nucleotide DNA duplexes containing either Watson-Crick, single, or tandem mismatch base pairs. We identified that TT/TT, C-C, and G-G mismatches exhibited enhanced europium (III) emission, suggesting loss of coordinating waters and direct coordination between the DNA and europium. NMR structural studies were used to provide additional details of sequence specific binding for these mismatches.
