Quinoline-based compounds have very important applications ranging from anti-cancer agents, anti-bacterials, antimalarial agents, etc. When used as antimalarials, quinoline-based drugs are believed to inhibit hemozoin formation by interacting with the prosthetic heme group. The mechanism of the inhibition process is a subject of scrutiny. We have prepared and characterized some synthetic heme models, ruthenium(II) porphyrin complexes, and have found them to react with quinoline-based antimalarial drugs to form heme-drug adducts. IR, UV-vis and 1HNMR spectral data of the adducts suggest Ru binding to the quinoline nitrogen of the antimalarial drugs. We have studied the electrochemical behavior of the heme-quinoline adducts obtained by cyclic voltammetry, which suggests porphyrin-centered oxidations. Our data gives insight on the role quinoline-based drugs play in inhibiting hemozoin formation.
