Self-assembling peptide materials have been studied widely for delivery of drugs and proteins. Here we present our efforts to deliver a biologically relevant signaling gas, H2S, from peptide-based hydrogels. Prepared from short peptides derivatized with S-aroylthiooximes—a functional group that releases H2S in response to thiols, the peptides self-assemble into one-dimensional nanofibers in aqueous solution. Upon addition of CaCl2, gels form at 1 wt% peptide concentration. These gels were tested as a treatment for intimal hyperplasia in human vein segments. Compared with controls, including a peptide incapable of H2S release, the gels reduced vascular smooth muscle cell proliferation while enhancing proliferation of endothelial cells. Combined, these effects limited intimal hyperplasia in human vein segments under static culture.
