<i>Escherichia coli</i> has shown to be a suitable and sustainable host of production of a variety of methylated natural products including psilocybin, vanillin, and certain anthocyanins. For these pathways to work efficiently, the <i>E. coli</i> host must provide enhanced production and regeneration rates for the releveant precursor and co-factor metabolites needed for these pathways. Here, we focus on optimizing the efficiency of the methylation reaction catalyzed by the S-adenosylmethionine (SAM) - dependent psilocybin methyltransferase (psiM). Efforts to improve methylation performance through appilication of transcriptional optimization, targeted gene deletions, and substrate supplementation techniques will be presented. We focus our efforts on SAM regeneration while also considering the complexities of the methionine pathway regulation on overall cellular health.
