Current understanding of how the mycobacterial cell envelope is remodeled during drug treatment is limited. Chemical tools that report on this phenotype while limiting cellular perturbation are critical to gaining more insight into this time-dependent phenomenon. We have developed a fluorogenic probe that reports on mycobacterial cell envelope assembly in real time. Using time-lapse microscopy, we revealed distinct spatial and temporal changes of mycobacterial lipids upon treatment with front-line antibiotics. Unique phenotypic profiles were observed upon differential antibiotic treatment thus providing a platform for simultaneously monitoring cell envelope construction and remodeling responses, which may provide insight into the development of resistance mechanisms.<br/>
