Metal-organic capsules offer fascinating opportunities to engineer host-guest interactions for a variety of applications. We sought to systematically study and optimize host-guest interactions in solution- and solid-states. Specifically, we focus on the influences from metal nodes and linker functional groups on the effective cavity, host-guest association, and guest exchange kinetics. These investigations led us to develop a metal-organic capsule-based <sup>129</sup>Xe magnetic resonance sensor, which is detectable at picomolar concentrations resulting from very favorable Xe exchange kinetics.
