N-pyridyl hydroxylamine scaffold is found in anti-bacterial, anti-viral, anti-osreoclastogenic and anti-cancer agents. N-Pyridyl hydroxylamine derivatives were prepared via copper-catalyzed cross-coupling of N,O-difunctionalized hydroxylamines and iodopyridines. Various traditional amino- and hydroxyl-protecting groups were tolerated and a total of 24 examples were synthesized in 28-91% yield. This method was employed towards the synthesis of a new class of hydroxylamine-based reactivators for nerve agent-poisoned acetylcholinesterase (AChE). A total 8 reactivators were prepared and the reactivation efficiency was evaluated against several organophosphorus nerve agents (OPNAs).
