IQGAP1 is a multi-domain protein that acts as a scaffold for multiple signaling pathways, especially in cancer cells where it is overexpressed. While IQGAP1 is known to generate the lipid messenger PI(3,4,5)P<sub>3</sub> by scaffolding the phosphoinositide kinases PIPKIs and PI3K in response to agonists such as EGF, the dynamics of this scaffold in intact, living cells remains unknown. Using imaging techniques including total internal reflection fluorescence microscopy (TIRF), the role of IQGAP1 in the PI3K signaling pathway. It was observed that upon stimulation with EGF, both EGFR and IQGAP1 accumulate along the cellular membrane of HeLa cells as well as in the lysosomes. By down-regulating IQGAP1 expression using siRNA, we see that IQGAP1 prevents EGFR accumulation along the membrane and promotes EGFR targeting to the lysosomes in the absence of EGF. These results indicate that IQGAP1 provides a protective effect to the membrane bound EGFR through scaffolding.<br/>
