Proteins comprise seven of the ten top-selling drugs on the market are proteins. However, most therapeutic proteins target extracellular receptors because the large size and hydrophilicity of most proteins prevent them from crossing the plasma membrane to access intracellular targets. Therefore, a great effort has been made by scientists to invent and improve intracellular protein delivery technologies, like lipid nanoparticles (LNPs). My project is to use Chloroalkane penetration assay (CAPA) to profile cell penetration of proteins and Lipid Nanoparticles. The first aim of my project is to use CAPA to design next-generation LNPs with combinatorial libraries. The second aim is to understand more about one important system, Cas9/LNP, with CAPA.
