G-rich sequences in the genome, including telomeres and near promoter regions, are shown to form four stranded quadruplex structures (G-DNA). Formation of these structures is shown to influence telomere biology and in the regulation of gene expression. Both DNA sequence and buffer conditions influence the morphology of these interesting structures that can fold into parallel, hybrid, and antiparallel forms. These factors also influence their interaction with small molecules through a range of binding modes such as intercalation, end stacking, or outside binding. Through the use of UV-Visible absorption spectroscopy, circular dichorism (CD) spectroscopy, and Fluorescence Emission spectroscopy, we explore the conformation of human telomere sequence Tel22, 5’-AGGG(TTAGGG)3-3’ in 50mMK+, 50mMNa+, and PEG and further investigate the effect of flanking sequences on the folding patterns of Tel22.
