Malate dehydrogenase (MDH) is an oxidoreductase enzyme that catalyzes the reversible conversion of malate and NAD<sup>+</sup>to oxaloacetate and NADH in both the cytosol and mitochondria. MDH contributes to the Malate-Aspartate Shuttle to maintain energy homeostasis across the mitochondrial membranes. The Konkle Lab previously found that the mitochondrial protein mitoNEET interacts with MDH enzymes through pull down assays with HepG2 cells and mouse liver lysates. The overall objective of this research is to understand how mitoNEET affects the enzyme kinetics and thermodynamics of MDH enzyme activity. The work presented here is the first step in this goal, optimizing the assay conditions for studying MDH enzyme kinetics and thermodynamics using UV-visible spectroscopy.
