N-acylhomoserine lactone (AHL) synthase in gram-negative bacteria make quorum sensing signals to regulate certain physiological activities such as virulence, biofilm formation, and toxin production. Since AHL synthase inhibitors would prevent the synthesis of quorum sensing signals, and thus interrupt microbial communication, these compounds have the potential to curb virulence in antimicrobial therapy. We propose that 3-oxo-alkyl-IACP probes will bind to a broad array of AHL enzymes, enabling the development of a high throughput assay for discovering AHL synthase inhibitors. We have synthesized few 3-oxo-alkyl-IACPs with varying linker chains to develop active site binders for Pseudomonas aeruginosa RhlI, Pantoea sterwartii EsaI, Burkholderia mallei BmaI1, and Yersinia pestis YspI AHL synthases. Fluorescence enhancement experiments reveal that the probes designed in this project do bind to AHL synthases. These initial encouraging results will enable us to assess binding affinity differences between native and non-native alkyl-chains to AHL synthase.
