Background：Food allergies are a major public health concern and represent an unmet clinical need. An individual can become allergic to any food. Food allergies often present as part of a constellation of allergic diseases, referred to as the atopic (or allergic) march. A number of theories have been proposed to explain the rising prevalence of allergic disease. Intestinal flora imbalance is considered to be one of the main causes.<br/>Method: A mouse allergy model of Wheat gluten were constructed, and stool samples of allergic mice were collected. And these samples were test by sequencing of the V3-V4 regions of the 16S rRNA gene. The tissues of small intestine were examined by HE strain. And the levels of OVA-sIgE in serum were measured by ELISA.<br/>Results: After sensitized, serum sIgE of allergic mice group was higher than the control group; HE staining showed that allergic mice group had more serious inflammation reaction than control group, such as the Inflammatory cells infiltration, the focal necrosis and shedding of the epithelial cells in intestinal villi; Sequencing results showed that the Firmicutes in control group were predominate, whereas the Proteobacteria were advantage bacterium in allergic mice group.<br/>Conclusions: Different microbes could affect the sensitivity of the inoculated mice on wheat gluten, which suggested abnormal intestinal flora may be the initiating factor in food allergies; the sensitivity of food allergy could be passed through the intestinal flora.
