There is an urgent and ever-persisting demand for the development of new antimicrobial drugs to treat serious and fatal illnesses. Pyrazoles are a class of compounds known to be privileged scaffolds for drug discovery. It was reported by Gautam et. al. that the pyrazole compound, 4-(5-(4-chlorophenyl)-3-methyl-1H-pyrazol-1-yl)-N-ethylaniline has both antibacterial and antifungal properties. In addition the pyrazole AR-12/OSU-03012 has demonstrated activity against a plethora of infectious agents, including bacteria, fungi, parasites, and viruses. Analogs of 4-(5-(4-chlorophenyl)-3-methyl-1H-pyrazol-1-yl)-N-ethylaniline were synthesized with the intent that they would also exhibit enhanced antibacterial activity if they possessed a glycine amide substituent, similar to that present in the AR-12/ OSU-03012. Approximately ten pyrazole analogs were synthesized, using the Knorr pyrazole synthesis, and fully characterized by 1H NMR, 13C NMR, TLC, HPLC, MP, and HRMS. The synthesized compounds were then evaluated for antibacterial activity using the Kirby-Bauer disk diffusion assays against E. coli. The antibacterial activity of the compounds was compared to the known fluoroquinolone antibiotic Norfloxacin.
